Sirtuin Pipeline

Sirtris is focused on discovering and developing proprietary, orally available, small molecule drugs with the potential to treat diseases associated with aging, including metabolic, inflammatory, neurodegenerative and cardiovascular diseases.

Working from the solid foundation established over several years, the Sirtris team has expanded upon the data from our proprietary formulation of resveratrol (SRT501) to focus on developing many novel small molecule activators of SIRT1. These new chemical entities or NCEs are novel compounds that are chemically and structurally unrelated to resveratrol and are more specific for the SIRT1 target with improved drug-like properties.

+SRT2104

SRT2104

In preclinical studies, SRT2104 demonstrated a lowering of blood glucose, reduced weight gain, increased energy expenditure, improved exercise tolerance and improved insulin sensitivity in several animal models of Type 2 Diabetes.

SRT2104 was Sirtris’ first NCE to enter human trials; the Phase I program in healthy volunteers demonstrated favorable safety and tolerability. We currently have under way multiple Phase I and II trials in patients with metabolic, inflammatory and cardiovascular diseases.

+SRT2379

SRT2379

Our second NCE to enter human clinic trials is SRT2379, which is chemically and structurally distinct from resveratrol and SRT2104. In preclinical studies, SRT2379 demonstrated efficacy in multiple disease models of diabetes and inflammation. SRT2379 is currently being evaluated in a Phase I clinical trial for safety, tolerability and pharmacokinetics.

+SRT501

SRT501

SRT501 is our proprietary formulation of resveratrol with improved bioavailability. In Phase I human clinical trials, SRT501 was found to be safe and well tolerated. We are currently testing SRT501 in a Phase II trial in patients with multiple myeloma; new patient enrollment has been suspended as of April 22, 2010 (see clinicalTrials.gov for updates).
